A cell culture platform for Cryptosporidium that enables long-term cultivation and new tools for the systematic investigation of its biology

چکیده
Cryptosporidium parasites are a major cause of diarrhoea that pose a particular threat to children in developing areas and immunocompromised individuals. Curative therapies and vaccines are lacking, mainly due to lack of a long-term culturing system of this parasite. Here, we show that COLO-680N cells infected with two different Cryptosporidium parvum strains produce sufficient infectious oocysts to infect subsequent cultures, showing a substantial fold increase in production, depending on the experiment, over the most optimistic HCT-8 models. Oocyst identity was confirmed using a variety of microscopic- and molecular-based methods. This culturing system will accelerate research on Cryptosporidium and the development of anti-Cryptosporidium drugs.

Identification of a Specific Pseudo attP Site for Phage PhiC31 Integrase in Bovine Genome
Background: PhiC31 integrase system provides a new platform in various felid of research, mainly in gene therapy and creation of transgenic animals. This system enables integration of exogenous DNA into preferred locations in mammalian genomes, which results in robust, long-term expression of the integrated transgene. Objectives: Identification of a novel pseudo attP site. Materials and Methods...

In the present study, an in vitro brain cell culture was developed from Persian sturgeon (Acipenser persicus). The tissues from anterior, middle and posterior regions of the brain were dissected, dispersed and cultivated separately in DMEM/F12 medium supplemented with 15% fetal bovine serum, antibiotic and antimycotic. The medium was changed every 3 days. The cells became confluent after about ...

متن کاملCultivation of Cryptosporidium in host free cells culture using Iraqi isolate
Cryptosporidiosis is a clinical disease, usually presenting as a gastro-enteritis-like syndrome, caused by infection with protozoan parasites of the Apicomplexan genus Cryptosporidium. Disease ranges in seriousness from mild to severe and signs and symptoms depend on the site of infection and nutritional and immune status of the host Cryptosporidium in vitro was first achieved in endodermal cel...
متن کاملMicroRNAs as a New Molecular Biomarker for Diagnosis and Prognosis of T-cell Acute Lymphoblastic Leukemia (T-ALL): A Systematic Review
MicroRNAs (miRNAs, miRs) are small endogenous non-coding RNAs that regulate the expression of protein-encoding genes at the post-transcriptional level. Several studies have described the role of miRNAs in T-cell acute lymphoblastic leukemia (T-ALL), including tumor suppressor and oncogenic miRNAs. Down-regulation of miRNA expression is a prominent feature of human malignancy. This down-regulati...
